The top large-letter sequence is from the Protein Family Database with the colored circles showing regions that correspond to CasA protein binding sites. The other three sequences, from bottom to top respectively, are the sequences for our ancestral reconstruction CasA protein, the current Thermus thermophilus CasA protein, and the consensus sequence of the two. The reconstructed gene only matched the Thermus thermophilus gene 25.9% of the time. This image shows how the matching regions are not random, but in fact correspond to the regions most important to the function of the protein.
