This morning we have fostered the bacteria with BBa_K741002 in M9 medium of 0mM, 500mM, 800mM, and 1000mM of glucose, as well as the bacteria with BBa_K774007 in LB of 0mM, 5mM, 10mM, 15mM, and 20mM. And in the afternoon, we have tested biobricks BBa_K741002 and BBa_K774007 using FCM (Flow Cytometry). For BBa_K741002, we saw the expected result: the bacteria in 0mM produced GFP in most strong intensity, and with the concentration of glucose increasing, the produced GFP was in lower and lower intensity. For BBa_K774007, we have seen the bacteria with no KNO3 has no RFP produced, and with the concentration of KNO3 increasing till 15mM, there was more and more RFP produced, but with the concentration of 20mM, the RFP produced less than that of 15mM. Since the FCM had not an exciting light around 590~600nm, so we didn’t get an obvious result from FCM, but the increase in RFP produced can be inferred from it.
