Biosensor

In order to construct aromatic biosensors and apply them to environmental detection, we selected XylS, HbpR, NahR from Pseudomonas, HcaR, MhpR, PaaX from E.coli. After obtaining their coding sequence via PCR or synthesis, we constructed an expression system, in which we link a constitutive Pc promoter with sensor’s coding sequence that regulates corresponding promoter located in the front of RBS-sfGFP fluorescence reporter. We test fluorescence strength to show induction ratio of each expression system at the presence of their inducers through ELIZA and cytometry. Based on these data, we selectively changed Pc and RBS strength, tuning expression of sensor and sfGFP respectively, to optimize the induction behavior. The fine-tuned expression system can be applied to multi-component analysis. Exposed to samples from environment, their On/Off behavior qualitatively reflect whether there are certain aromatic compounds.
