The plate containing ligation product BBa_K1085014-BBa_K1085018 showed around a hundreds of colonies.
Colony PCR (NEB Master Mix) was performed on 16 colonies using F-hy_spank-NheI and HM14 primers (annealing temperature 70°C).
The samples were checked on agarose gel 0.8%.

Colony 13 from plate BBa_K1085014-BBa_K1085018 showed to be a positive candidate (expected band ~2400 bps) and was inoculated in LB + Amp (Sander).
