96-Well Plate Assay Obtain 96-Well Plate. 1. Pipette 260 ul of the overnight culture prepared according to the general protocol to each well being tested and cover with a breathable plate seal. 2. Set the E. colight app with the appropriate conditions. 3. Orient the 96-well plate on the tablet such that the wells line up with the appropriate light source on the tablet and secure with tape. 4. Incubate tablet and 96-well plate on an orbital shaker at 240 RPM for 16 hours at 37C. 5. Follow the Fluorescent Analysis Protocol.
