Overview
Many methods have been found to regulate metabolism expression so far , such as RNA interference(RNAi) and engineered DNA-binding proteins(such as zinc finger protein) are usually used in relevant study. | |
However, these methods are not easy enough for one carrying out a result. Or sometimes, due to their limitation, we can only use them in very specific creatures. Usually, a large quantity of experiments and data need to be determined to make one pathway clear, which cost a lot. | |
Fortunately, many new findings make us can build an easy system to regulate metabolism system. We assemble the light sensor with sgRNA in one part to construct the system. Light sensors are widely used in engineered biosynthetic parts, for their sensibility and adjustable intensity. Thus, we can simply use kinds of light sensor control the expression of sgRNA. The sgRNA is another fascinating tool, which can bind target sequence by mutating its changeable region. Flexible function makes it qualified for most creatures, not only protokaryon, but also eukaryon. Moreover, this novel device provides us a promising way of genome-scale metabolism regulation. Combining the light sensor with our system, parameters determination of multiple genes expression becomes more predictable and precise. | |
In our project, we use three kinds of light sensors meeting our demands that the absorption wavelength of each has the minimum overlapping area to build a prototype system. In future, we want to apply our device on multiple steps in metabolism networks. By accomplishing this goal, more acceptable light sensors should be found. | |
