We were planning to perform Western Blot analysis in order to check the specificity of binding between CEA protein and DNA aptamers from SELEX 7th and 8th cycles. However, we found out that the minimum concentration of the protein for Western Blot is 0.1mg/ml, and the optimum concentration is in the range of 1-5mg/ml. It means that if we take 1mg/ml, the concentration of our protein needs to be around 5.6 uM. In our lab, we have only 38.9 nM concentration of CEA. Therefore, Western Blot analysis could not be performed.
