== 1 Project description ==<br />
We try to design a kind of beverage, containing engineered strains, to resist alcohol. Alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) can be expressed in the strains and after lysis, the two enzymes can transfer ethanol into acetic acid in the stomach. Then drunkenness can be relieved. This model aims at describing how the concentration of substrates changing under the function of our enzymes. And an instruction is given to help consumers to decide how much beverage they need according how much alcohol they drink.<br />

We mainly build a group of enzyme kinetic equations to describe the enzyme catalysis reactions. With this model, we can know how much ethanol can be transferred successfully with a certain amount of enzymes.<br />

Because of the gastric motility, the maximum of time is 4 hours. And the volume of gastric contents is about 1L, the value of a half full adult. Because we aim to improve the enzyme activity at pH2.0 to a natural or higher level, so the k1to k8 are cited from the reported data[1][2]. The data are got at the optimal conditions. <br />

We mainly use Matlab to solve the ordinary differential equations. The analysis is based on the Runge-Kutta method of 4 step algorithm. <br />
A bottle of 75cl wine contains about 1.32mol ethanol. With the activity of 0.02uM ADH and 0.4uM ALDH, the concentrations of each substrate can be shown as figures. We can see that the ethanol can be degraded within about one minute, which indicates the effectiveness of our beverage.<br />

At the same time two thirds of people don’t believe all kinds of health products which can de-alcoholic or protect the liver. Ninety percent of people want to reduce the harm of drinking to health and they also believe that the biotechnology can metabolize alcohol before it harms the health. Almost eight over ten will take a try if there is a magic drinking with no side effect. In short, from our survey, people will accept the idea of a magic drinking that can reduce the harm of the alcohol. <br />

On the evening of September 7th, we offered an enjoyable introduction to the freshmen who just entered China Agricultural University (CAU) this semester, about our iGEM. <br />

On the evening of September 7th, we offered an enjoyable introduction to the freshmen who just entered China Agricultural University (CAU) this semester, about our iGEM. <br />

Some students brought out their hope and goals that once they joined in such a team they wanted to achieve. And many of them are attracted by the creativity creative by the iGEMers.<br />

The youngest member also shared her ideas about the experiments and the experience in the lab of iGEMers. Unfamiliar as a new one may be with the activities, they can make it with the help from those who are experienced. <br />

After the introduction, we made a survey on how the students thought of the drink with alcoholic sobering effects, which is the direction we head for. It was just a part of our research for more people to know what we interested in.<br />

After showing the things that we majored in and the goal we chased after, the team appealed to plenty of the students who were thirsty for some more information about the team. With their enthusiasmaroused, some of them showed ambitions that they wanted a try. <br />

In order to understand the ideas what people think about drinking, we did a significant research in which there were seven questions. Through four ways of researching, we recycled 650 questionnaires and obtained some conclusions as following. Half of people wouldn’t drink frequently. However, more than two thirds of people are not bothered by drinking or alcohol. At the same time two thirds of people don’t believe all kinds of health products which can de-alcoholic or protect the liver. Ninety percent of people want to reduce the harm of drinking to health and they also believe that the biotechnology can metabolize alcohol before it harms the health. Almost eight over ten will take a try if there is a magic drinking with no side effect. In short, from our survey, people will accept the idea of a magic drinking that can reduce the harm of the alcohol. <br />

== About questionnaire ==<br />

<br />
In order to understand the ideas what people think about drinking, we did a significant research in which there were seven questions. Through four ways of researching, we recycled 650 questionnaires and obtained some conclusions as following. Half of people wouldn’t drink frequently. However, more than two thirds of people are not bothered by drinking or alcohol. At the same time two thirds of people don’t believe all kinds of health products which can de-alcoholic or protect the liver. Ninety percent of people want to reduce the harm of drinking to health and they also believe that the biotechnology can metabolize alcohol before it harms the health. Almost eight over ten will take a try if there is a magic drinking with no side effect. In short, from our survey, people will accept the idea of a magic drinking that can reduce the harm of the alcohol. <br />

On the evening of September 7th, we offered an enjoyable introduction to the freshmen who just entered China Agricultural University (CAU) this semester, about our iGEM. <br />
<br />

Some students brought out their hope and goals that once they joined in such a team they wanted to achieve. And many of them are attracted by the creativity creative by the iGEMers.<br />
<br />
The youngest member also shared her ideas about the experiments and the experience in the lab of iGEMers. Unfamiliar as a new one may be with the activities, they can make it with the help from those who are experienced. <br />

After the introduction, we made a survey on how the students thought of the drink with alcoholic sobering effects, which is the direction we head for. It was just a part of our research for more people to know what we interested in.<br />
<br />
After showing the things that we majored in and the goal we chased after, the team appealed to plenty of the students who were thirsty for some more information about the team. With their enthusiasmaroused, some of them showed ambitions that they wanted a try. <br />

We invited the team leader from Wuhan University to our lab. Both teams introduced the projects and gave some suggestions to each other. It was a good experience to share the enthusiasm towards iGEM. <br />

On the evening of September 7th, we offered an enjoyable introduction to the freshmen who just entered China Agricultural University (CAU) this semester, about our iGEM. <br />

Some students brought out their hope and goals that once they joined in such a team they wanted to achieve. And many of them are attracted by the creativity creative by the iGEMers.<br />

The youngest member also shared her ideas about the experiments and the experience in the lab of iGEMers. Unfamiliar as a new one may be with the activities, they can make it with the help from those who are experienced. <br />

After the introduction, we made a survey on how the students thought of the drink with alcoholic sobering effects, which is the direction we head for. It was just a part of our research for more people to know what we interested in.<br />
<br />
After showing the things that we majored in and the goal we chased after, the team appealed to plenty of the students who were thirsty for some more information about the team. With their enthusiasmaroused, some of them showed ambitions that they wanted a try. <br />

We invited the team leader from Wuhan University to our lab. Both teams introduced the projects and gave some suggestions to each other. It was a good experience to share the enthusiasm towards iGEM. <br />

== About questionnaire ==<br />
In order to understand the ideas what people think about drinking, we did a significant research in which there were seven questions. Through four ways of researching, we recycled 650 questionnaires and obtained some conclusions as following. Half of people wouldn’t drink frequently. However, more than two thirds of people are not bothered by drinking or alcohol. At the same time two thirds of people don’t believe all kinds of health products which can de-alcoholic or protect the liver. Ninety percent of people want to reduce the harm of drinking to health and they also believe that the biotechnology can metabolize alcohol before it harms the health. Almost eight over ten will take a try if there is a magic drinking with no side effect. In short, from our survey, people will accept the idea of a magic drinking that can reduce the harm of the alcohol. <br />

== 1 Project description ==<br />
We try to design a kind of beverage, containing engineered strains, to resist alcohol. Alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) can be expressed in the strains and after lysis, the two enzymes can transfer ethanol into acetic acid in the stomach. Then drunkenness can be relieved. This model aims at describing how the concentration of substrates changing under the function of our enzymes. And an instruction is given to help consumers to decide how much beverage they need according how much alcohol they drink.<br />

We mainly build a group of enzyme kinetic equations to describe the enzyme catalysis reactions. With this model, we can know how much ethanol can be transferred successfully with a certain amount of enzymes.<br />

Because of the gastric motility, the maximum of time is 4 hours. And the volume of gastric contents is about 1L, the value of a half full adult. Because we aim to improve the enzyme activity at pH2.0 to a natural or higher level, so the k1to k8 are cited from the reported data[1][2]. The data are got at the optimal conditions. <br />

==== 2.4 Model solution ====<br />
We mainly use Matlab to solve the ordinary differential equations. The analysis is based on the Runge-Kutta method of 4 step algorithm. <br />
A bottle of 75cl wine contains about 1.32mol ethanol. With the activity of 0.02uM ADH and 0.4uM ALDH, the concentrations of each substrate can be shown as figures. We can see that the ethanol can be degraded within about one minute, which indicates the effectiveness of our beverage.<br />

Consumers should drink the alcohol-toxic beverage firstly and then drink alcohol. The time interval should be no longer than 4 hours and the shorter, the better. According to the results of the model, consumers can choose how much beverage they need according to how much alcohol they drink.<br />

However to human’s surprise, almost all forms of life share the same basic components. There are rules to connect simple form and complex apparent just like the game of go .I believe all those rules will come out in same simple way. To pursue those rules is my lifelong career and nothing could make me more excited. I graduate from CAU and my specialized field is calculation biology. Hoping my effort could partly contribute to CAU iGem team's achievement. <br />

Everybody here is excellent and special. For my part, acid resistance domestication of E.coli(BL21) was the main work. <br />

Life is simple and life is complex .Creatures are everywhere on earth from South Pole to the bottom of Pacific, in varieties forms from mycoplasma to mammal. However to human’s surprise, almost all forms of life share the same basic components. There are rules to connect simple form and complex apparent just like the game of go .I believe all those rules will come out in same simple way. To pursue those rules is my lifelong career and nothing could make me more excited. I graduate from CAU and my specialized field is calculation biology. Hoping my effort could partly contribute to CAU iGem team's achievement. <br />

I focus on the work relating to molecular cloning operation, such as construction of various basic plasmid backbones. And to tell the truth, I’m also a completest to some extent, which really exhausts me sometimes. Very fortunately, I’ve got so many good friends since I joined this team. Everybody here is excellent and special. For my part, acid resistance domestication of E.coli(BL21) was the main work. <br />
