Making LB Plates

Prepare Required Solutions

Prepare Plates
  • Gather sleeves of petri dishes
    • Keep the plastic sleeves to place plates into
    • Label the petri dishes with date and antibiotic
  • Place the plates open in a line when done with labeling
Prepare Solution
  • Use 2 L Erlenmeyer Flask for the Agar solution
  • Add dry materials and water to flask
  • Add magnetic stir bar and stir the media
  • Cover the top of the flask loosely with aluminum foil
  • Autoclave the media for 30 minutes on a fluid cycle
Pouring the Solution
  • Let the solution cool to ~60C
  • Test this with hands (when hand can stay on flask for more than 5 seconds)
  • Use water bath
  • Pour the agar into each plate
    • Go down the line, recapping after each plate is poured
    • Use sterile technique
    • Fill each plate to about half-full of agar
    • If there are bubbles, try to pop them by passing plate over a flame
    • Add antibiotics to solution if needed
  • Let the plates to dry and cool overnight
    • Let the plates solidify right side up
    • Flip the agar side on top to keep condensation off of the gel when solid
    • Leave out on lab bench
    • Next day, store the plates in original sleeves in 4C
