Low Iron Medium (LIM)

Reagents

6.6 g Yeast nitrogen base
0.83 g Synthetic complete drop out mix
55.0 mg Adenine sulfate
1 mM EDTA
16.6 g Agar
to 1000 mL Aqua dest.

 

Procedure

For plates add agar, for liquid medium leave out agar.
Mix all reagents except for agar in an autoclavable flask and fill up to 1000 mL with Aqua dest. Adjust pH to 5.6. Add agar and autoclave mixture. After autoclaving, let cool down a bit and add antibiotics if medium is required for selective purposes. Perform next steps at laminar flow cabinet! Cast plates in petri dishes (approx. 20 mL per plate) while medium is still warm. Store plates at 4 °C.
