Making LB and TB Media


TB media: ( Don’t use Capsules, use Powder)
1. Get large glass bottle and the cap. LABEL what is inside, date, and initials.
2. Get plastic weighboat, put on scale, turn on the scale, and tare the scale. (Use Postage Scale - The big ones)
3. Weigh TB powder for 500ml (named: Terrific Broth by EMD, blue capped, doesn’t have glycerol), amount to use is written in the instructions, follow closely. Put it inside the bottle.
4. Bring back bottle to iGem lab and add 50% glycerol, double the amount needed as stated from instruction on the bottle when using 50%.
5. Add diH2O until 500ml mark.
6. Place lids loosely on top and foil the tops. Put autoclave tape on it.
7. Label. Autoclave it. (have someone show you if it is your first time)

LB media:
(Don’t use LB made from capsules for miniprep)
1. Get large glass bottle and the cap. LABEL what is inside, date, and initials.
2. 1 large pill of LB medium per 125 ml, put inside of bottle
3. Add diH2O until 250ml mark
4. Place lids loosely on top and foil the tops. Put autoclave tape on it
5. Label. Autoclave
