Today we performed TOPO TA cloning of our PCR products of DNA aptamers from SELEX cycle 8. For this procedure we used pGEMTII Vector. The following steps were done:
1. Ligation reaction by inserting our DNAs into pGEM vector
2. Transformation of pGEM vector into JM109 High Efficiency Competent Cells
3. Plating cells on LB/Ampicillin/IPTG/X-Gal plates
