From 2013.igem.org
TOPO TA cloning of our PCR products of DNA aptamers from SELEX cycle 8 was done. For this procedure we used pGEMTII Vector. The following steps were done:
Ligation reaction by inserting our DNAs into pGEM vector
Transformation of pGEM vector into JM109 High Efficiency Competent Cells
Plating cells on LB/Ampicillin/IPTG/X-Gal plates
In addition control and test samples were innoculated into LB broth for overnight incubation.
