DOT ELISA (see protocols for details) was done to see the binding between anti-CEA antibodies and biotynilated CEA. The results showed very slight binding at very high concentrations.

Since it is unknown if our biotynilated CEA has the proper size of 180 kDa, it was decided to check its size with SDS PAGE (see protocols for details). The results of SDS PAGE showed that biotynilated CEA, that we have, does not have proper size. Indeed, it is less that 180 kDa. Furthermore, it contains several subunits, thus the gel showed double bands for this protein.
