We had a meeting with our advisors, and today all members of the team were present (Alexander joined us today). Mukhtar made a presentation about what we did during the last week. We had also discussed some new approaches that can be used in our project, in particular, about using the parts, which will help to express streptavidin on the surface of the cell. We were discussing the following parts: BBa_J36847 and BBa_K215250. We decided to search more on this topic and to meet tomorrow for brainstorming.

The good news for us today was that we have received our kit!

After that, we went to the lab to prepare the gel for running dsDNAs from SELEX 7 (one is extracted from the gel and the other is from PCR). After running our products we could see the bands only for dsDNA from PCR. We cut the band of 80 bp from the gel and left it at -20degC for further extraction and amplification. Finally, we discussed the results of EMSA, and decided to increase the concentration of ssDNA and to repeat the experiment.
