After confirming, our biobrick has been sent back, so we started to construct 11 plasmid, which were named 00, 01, 02, 03, 04, 05, 06, 07, 08, 09, zeo. In order to make sure we could make our biobricks successfully, we decided to use the plasmid from BBa_K774007, BBa_K741002, and iGem. Besides, we had to cut our biobricks out of its original plasmid. Disappointedly, after running glue, it seemed like we didn’t cut it out, but there really some in the colloid, so we just incise what we sure, in which way, we only got it in a very low concentration. Then we continued to do the linking and transformation. Just after transformation, we fostered the bacteria for 40 minutes, but we didn’t see any bacteria grew up in LB. It’s so strange, and we started to suspect whether the plasmid has been transformed in. Since we had no time waiting to see the result of this transformation, we then did another transformation again along with the first one. We had finished these all at 6:20 in the morning of 9-15-2013.
