After a happy journey to Hong Kong, we were absorbed in our experiments again. What was a hit to us was that there were no bacteria community on the plate again, whether the xylose was added or not, which frustrated us greatly for the second time. We assumed a good result after the failure last time. Then we began searching the reasons and we scan the web page introducing this promoter, we found the bacteria were cultured on the M9 culture. We guessed this was the reason, then we prepared some M9 culture.
