The situation became even worse, to be honest, we were worried about schedule of our whole project, for the time left was not that much and we got no progress. Again there were no bacteria communities on the plate, we got into an embarrassing situation. For the consideration of the whole project, we began finding new biobrick and repeated the procedure yesterday.
