Flourescence
Green fluorescent protein (GFP) and monomeric red fluorescent protein (mRFP) are frequently used as reporter markers for the characterization of gene expression. In developing our fungal toolkit, we cloned GFP and mRFP downstream of numerous promoters to quantify promoter strength in the style of Toews et al, facilitating future genetic work with basidiomycetes [1]. The fungal promoters employed included T7, ''PtrpC'', and A. nidulans PgpdA.We are also collaborating with Wageningen's iGEM team to characterize their team's actin-GFP fusion construct in our chassis organisms.
