4.4 Glycerol Stocks


I) Purpose

Create glycerol stocks to store viable bacteria for use.

II) Expected Outcome

Bacteria that can be retrieved from the freezer and used at any time.

III) Reagants Used

W3110 liquid culture
BL21 Liquid culture
40% Glycerol


IV) Actual Procedure

Placed .5 mL cultures of liquid bacteria from the cultures on 4.3 W3110 Liquid Culture into cryo vials.
Added 500 mL 40% glycerol.
Vortexed for 10 seconds.
Put in the -80 C freezer for storage.

V) Results

Bacteria will be stored and available for use until no longer viable or contaminated.
