Summary:
In this summer, we construct a new alkane sensing system AlkR-PalkM, all the achievements are based on this system.
- Based on the system of AlkR-PalkM , we use this to build an Alk-Sensor based on the system of AlkR-PalkM which can detect the alkane which is produced by engineered E. coli.
- We characterized the Alk-Sensor of different aspects.
- Based on Alk-Sensor, we constructed Alk-Selector for direct evolution
- We used Alk-Selector to optimize the function of NPDC.
Future work:
All parts are also not perfect, needed to be optimized.
- It may be necessary to mutate PalkM to reduce the leakage of the system.
- Different reporter needed to be tried to get different relationship between input and output.
- Mutate AlkR may promote the interaction between AlkR and Alkane, and improve the function of AlkR-PalkM system.
- Addition of gene related to biosufactant biosynthesis may increasing the sensing range of the alkane
- For biofuel synthetic, Alk-Selector provide the more opportunities to engineered the strain. Many method related to build a library can be used in this field with the help of Alk-Selector.
