all cultures of yeast left for prorulation 5 days ago were not appropriate for further manipulations. After observing each sample microscopically, we could see either non-meiotic yeast cells or nothing. therefore the procedure was done from the beginning. the loopful of culture from glycerol stock was innoculated into each YPD containing tube and left for overnight incubation.
