Two colonies are chosen and inoculated in LB medium with Cm to higher the amount of the Pdes-cheY construct. Did a miniprep on the samples.
Inoculations of ΔDesΔCheY and WT bacillus strains in minimal medium.
Did a gel purification of CheC up and CheC down. Did restriction analysis of CheC up, CheC down and Spec. Ligation of CheC. Transformation of CheC in the ΔDesΔCheY strain.
