XMU-iGEM 2013

Project

By constructing robust circuits in E.coli, we built a gene network capable of synchronizing genetic oscillations in multiple levels (mainly AHL in liquid phase& H2O2 in gas). DNA agarose electrophoresis & SDS-page was used to confirm our construction.
In order to detect the oscillate in real time, we designed a liquid crystal display (LCD)-like microfluidic array which separates the bacteria in colonies.

Software

We coded two software by Matlab related our experiment.
First one is Gene OS, the model of oscillation which simulates our circuit and compares various results with different initial conditions. This gave us a deeper understanding about the mechanism.
The other one is ImageMe which offers batch processing function and reduces much of time in analyzing the microfluidic images.

Society & Safety

Nowadays, more and more people tend to build an environmentally friendly society and take more seriously about the safety issues of wet lab.
Our team used non-pathogenic E. coli strains to construct our circuit and gathered all culture fluid and culture medium with cells for unified treatment.
A series of other measurements were taken to reach the safety requirements.
