5.6 Phage From Lysate
I) Purpose
- Separate Phage from bacterial lysate to store viable phage for extended periods of time.
II) Expected Outcome
- Viable phage, easily stored.
III) Materials Used
- 2 mL eppendorf tubes (Qty 20)
- 5 mL cultures of BL 21 (infected with T7 phage for 24 hours) and W3110 (infected with T4 phage for 7 hours)
IV) Actual Procedure
- Separated the 5 mL cultures into 5 eppendorfs, 1 mL in each.
- Centrifuged at 3050 rpm for 5 min
- Gently extracted the supernatant and placed into clean eppendorf tubes.
V) Results
- The phage were still viable and are stable when stored in the fridge.
