  • Digestion of pieces: with the 3 hour method
  • Backbone (BB) - used the pATBS06 plasmid from before
  • Enzymes: NdeI and XhoI
  • Buffer 4
  • Expected length: 2200bp
  • There were a few bands on the gel, but I cut out the 2200bp one.
  • OmpR
  • Enzymes: NdeI and BamHI
  • Buffer 3
  • Expected length: about 720bp
  • cut out the piece
  • Aer/EnvZ(piece that was messed up. Just to see if it would actually work)
  • Enzymes: BglII and XhoI
  • Buffer 3
  • Expected length: about 1490bp
  • Didn’t show up on the gel because the piece was messed up
