5.6 Phage From Lysate


I) Purpose

Separate Phage from bacterial lysate to store viable phage for extended periods of time.

II) Expected Outcome

Viable phage, easily stored.

III) Materials Used

2 mL eppendorf tubes (Qty 20)
5 mL cultures of BL 21 (infected with T7 phage for 24 hours) and W3110 (infected with T4 phage for 7 hours)


IV) Actual Procedure

Separated the 5 mL cultures into 5 eppendorfs, 1 mL in each.
Centrifuged at 3050 rpm for 5 min
Gently extracted the supernatant and placed into clean eppendorf tubes.

V) Results

The phage were still viable and are stable when stored in the fridge.
