Gibson Assembly

The Gibson assembly binds several DNA fragments with complementary ends. To realize a Gibson Assembly, we need put together 3 enzymes with different functions: an exo-nucléase in 5’, a polymerase and a ligase.

1. Put in a tube 15µL of mix Gibson

2. Add DNA fragments in molar relation

3. Add water until 20µL

4. Incubate at 50°C during 1h30 in PCR
