Biological Construction

Riboregulation System

In order to better control the expression of the recombinases, a ribo regulation system have been add before the recombinases genes.

How does it work ?

This is the basic system : Two promoters each followed by a RNA sequence

The red and the blue regions are regions which code for little RNA sequences.


When P2 is activated and not P1, the blue sequence will be transcripted. This blue sequence possess a similarity with the rbs sequence. So when transcripted the blue sequence will confine the rbs and prevent the recombinase from being translated.

When P1 and P2 are activated, both sequences (the red and the blue one) are transcripted.
The red RNA sequence possess a stronger similiraty with the blue one. When transcripted the red and the blue sequences will match together and release the rbs to permit the translation of the recombinase
